Micropropagation of Chrysanthemum (chrysanthemum Morifolium) Using Shoot Tip as Explant
نویسنده
چکیده
Chrysanthemum has been cultivated for more than 2000 years and today, it is the world’s second most economically important floricultural crop following the rose (Teixeira da silva, 2003). Due to high popularity and demand for chrysanthemum it has become one of the first commercial targets for Micro propagation and thus tissue culture can be utilized for its large scale production. Regeneration of chrysanthemum plantlets was obtained by shoot tips as explant with different concentrations and combinations of auxins (IAA, IBA) and cytokinin (Kinetin), for the formation of micro-shoots, which were sub-cultured for development of roots. Fully developed plantlets were successfully transferred to suitable growing media for acclimatization. Among the various treatments for shoot regeneration, MS medium with Kinetin 3.0 mg l + IAA 2.0 mg l was found to be the best treatment combination as it produced 67.82 per cent success in 17.91 days. For root production, MS medium supplemented with IBA 1.0mg l was found to be the best treatment as it produced 82.12 per cent success in 19.19 days.

Flowering retardation by high temperature in chrysanthemums: involvement of FLOWERING LOCUS T-like 3 gene repression
Flowering time of the short-day plant Chrysanthemum morifolium is largely dependent upon daylength, but it is also distinctly influenced by other environmental factors. Flowering is delayed by summer heat. Here, the underlying basis for this phenomenon was investigated. Heat-induced flowering retardation occurred similarly in C. morifolium and C. seticuspe, a wild-type diploid chrysanthemum. In...

An efficient in vitro plant regeneration protocol was developed for Chrysanthemum morifolium, an ornamental plant. Nodal segments were used as explants in the present experiment. The explants were cultured onto Murashige & Skoog’s (MS) nutrient medium containing plant growth regulators (cytokinins or auxins) with various combinations and concentrations for the study. Maximum percentage (93.33%)...
متن کاملCallus Induction and Plant Regeneration of Chrysanthemum morifolium and C. coccineum via Direct and Indirect Organogenesis and Genetic Fidelity Analysis Using IRAP, ISSR and SCoT Molecular Markers
In vitro propagation of C. morifolium cv. ‘Homa’ and cv. ‘Delkash’ and wild C. coccineum via direct and indirect organogenesis and somatic embryogenesis were investigated. BAP at 0, 1, 2 and 3 mg l-1 or NAA at 0, 0.05, 0.1 and 0.2 mg l-1 concentrations were used to induce direct and indirect organogenesis of shoot tip explants. To study the callu...
متن کاملGeneration of Gene-Edited Chrysanthemum morifolium Using Multicopy Transgenes as Targets and Markers.
The most widely used gene editing technology-the CRISPR/Cas9 system-employs a bacterial monomeric DNA endonuclease known as clustered regularly interspaced short palindromic repeats (CRISPR)-associated protein 9 (Cas9) and single-guide RNA (sgRNA) that directs Cas9 to a complementary target DNA. However, introducing mutations into higher polyploid plant species, especially for species without g...
متن کاملFlower Colour Modification of Chrysanthemum by Suppression of F3'H and Overexpression of the Exogenous Senecio cruentus F3'5'H Gene
Chrysanthemum (Chrysanthemum × morifolium) is one of the most important ornamental plants in the world. They are typically used as cut flowers or potted plants. Chrysanthemum can exhibit red, purple, pink, yellow and white flowers, but lack bright red and blue flowers. In this study, we identified two chrysanthemum cultivars, C × morifolium 'LPi' and C × morifolium 'LPu', that only accumulate f...
